Abstract:
Cytogenetic investigations on peripheral blood lymphocytes of 65 workers exposed to Benzene in offset printing press were undertaken. These were compared with an equal number of occupationally unexposed and matched controls in relation to age, sex, smoking and drinking habits. The chromosomal aberrations (CA), sister chromatid exchanges (SCE), satellite associations and micronuclei (MN), were analysed. All the parameters showed a significant increase (P<0.01) in the exposed sample compared with the controls viz CA, 0.88-2.78, SCE, 3.72-9.08, SA, 4.53- 11.75 and MN, 0.45-0.57. A negative correlation has been established when comparing the data on sister chromatid exchange frequency and DNA repair capabilities of lymphocytes in patients with non-specific inflammatory pulmonary diseases. The role of DNA repair mechanisms in the alteration of sister chromatid exchange frequency in lymphocytes is discussed.

The genotoxic effect of occupational exposure of 20 nurses who handled cytostatic drugs in medical oncology and haematology units was evaluated by micronucleus and sister chromatide exchange test. The duration of employment in the units and of exposure to cytostatics ranged from 1 to 31 years. The exposed nurses manifested an increase in cells with micronuclei as compared to the control group (P < 0.05). Nurses exposed to cytostatic drugs for 20–31 years showed a higher frequency of micronuclei (P < 0.05), whereas there was no difference in frequencies between the control group and the group exposed for 1–14 years (P > 0.05). The influence of the exposure period proved to be a significant parameter for the micronucleus test. No statistically significant differences were observed in sister chromatid exchange (P > 0.05). Data are presented establishing a direct correlation between 3-aminobenzamide-induced sister chromatid exchange (SCE) frequency and the level of bromodeoxyuridine (BrdU) incorporated into DNA. In addition, it is shown that most of the SCEs are induced in the second cell cycle, in which BrdU-containing DNA is used as the template for replication.

An in vivo system for differentially stained sister chromatids by incorporating 5' Bromo 2' deoxyuridine at two consecutive round of DNA replication has been developed in C. punctatus. The base line developed frequency of sister chromatid exchanges (SCEs) was found to be 0.038 SCE/chromosome. This low baseline frequency of SCEs could be useful in detecting genotoxicity of pollutants in aquatic medium.
