Abstract:
Despite of recent advances on acknowledge of hepatitis B virus (HBV) structure and biology, little is known about the morphogenesis and release of virions. In this study, the ultrastructural analysis of HBV components in liver biopsies from chronically HBV-infected patients disclosed complex assembly and morphogenesis pathways for HBV. HBV core (HBcAg) and surface (HBsAg) antigens were specifically identified in all liver biopsies from HVB-infected patients. HBcAg containing core-like particles 24-28 nm in diameter were observed both in nucleus and cytoplasm of hepatocytes. Dane’s-like particles were detected either budding to or into the lumen of ER close to HBsAg containing tubular structures. Besides, Dane’s-like particles were detected in different vesicular compartments resembling multivesicular endosomes. Other kind of enveloped HBV-like particles similar to the previously described cobra-shaped and horn-shaped particles were also observed in hepatocytes. Some of these particles were connected to the vesicle membrane through a stalk 20-22 nm in diameter. On the other hand, spherical subviral particles (SVP) were frequently observed in cytoplasmic vesicles. Moreover, a minor proportion of enveloped HBV-like particles budding through the plasma membrane to the extracellular space and bile canaliculi were detected. Interestingly, Dane’s-like particles in the bile canaliculi and entering into ductular-like cells were shown. Strikingly, Dane’s-like particles close to tubular structures and specific immunolabeling for HBcAg both in cytoplasm and nuclei of stellate-like cells were detected. Remarkably, HVB-like particles appeared entering hepatocytes from large cytoplasmic processes of fibrocytes raising the interesting possibility of a cell to cell passage of HBV through direct transcellular migration.Keywords:

A comparative ultrastructural study was carried out on several species of Actinomyces and related Gram‐positive rods including Arachnia, Bacterionema, Rothia and Leptotrichia . A total of 52 well characterized strains were examined by transmission electron microscopy. Particular attention was paid to the ultrastructure of the cell periphery, that is the plasma membrane, periplasmic space, the cell wall per se and extramural structures, including surface “fuzz”. In addition, the ultrastructure of various features of the cytoplasm were also examined. A good correlation appeared to exist between certain ultrastructural characteristics of the microorganisms and their taxonomic position as determined by other criteria. It should be noted, however, that the ultrastructure of certain strains differed markedly from that of the remaining strains of the species. This observation raises some doubt on the appropriateness of the current classification for these particular strains. The ultrastructural features of the cell periphery were found to be particularly stable for all strains grown under standard conditions. For this reason, ultrastructural features of the cell wall and associated structures are probably a more reliable source of morphological criteria for identification purposes than the ultrastructural characteristics of the cytoplasmic components which tend to be more variable. The results suggest that certain ultrastructural features are useful criteria for the identification and classification of these Gram‐positive rods.

Objective:To explore the effects of IF and ultrastructure of rat's smooth muscle(SM) by swimming exercises,so as to provide some morphological basis of the changes of physiology and pathology by the exercise-induced change of smooth muscle.Methods:After exercises of EST,rat's smooth muscle were cut off.These specimens were treated respectively with regular procedures of paraffin sections and transmission electron microscope(TEM).The paraffin sections were stained by Des and Vim immunhistochemistry,their mean optics density and percentage of stained areas were observed and calculated.The ultrastructure of muscles were observed by TEM.Resuilts:In 8-week EST,Des,Vim and ultrastructure in SM of LIT had no significant changes compared with ETC.In MIT,Des and Vim of SMM increased obviously and the ultrastructure of SMM had adaptable changes.In HIT,Des and Vim decreased remarkably.At the same time,the injury changes were found in ultrastructure of SM.Conclusion:Swimming exercises with different intensity endurance would have different effects on IF and ultrastructure of SMM.

The aim of the study was to examine the ultrastructure of Anthracoidea elynae ustilospores isolated from Kobresia myosuroides (Vill.) Fiori plant ovaries, harvested in the Bucegi Mountains, Romania. Samples examination was performed using scanning (SEM) and transmission (TEM) electron microscopy. The results showed that A. elynae ustilospores had a dynamic ultrastructure, because their three-layered cell wall, nucleus shape, lipid and glycogen accumulations in the cytoplasm changed at each developmental stage. In conclusion, according to the ultrastructural changes, A. elynae ustilospores development may be divided into three stages.

Weinreb first reported the results of transmission electron microscopy observation on ultrastructure of goldfish erythrocyte in 1963. In recent years, there are few reports about the ultrastructure of blood cells in fish For further understanding the fine structure of mature erythrocyte in teleosts, using goldfish (Carassius auratus) as the experimental material We made an observation of the morphology of its mature erythrocyte by means of scanning electron microscopy as well as a study on the ultrastructure by freeze etching technique. Since there are none of reports concerning the study on erythrocyte in fish by using freeze etching technique, the present note will provide here a practical method and the data of studying about ultrastructure of erythrocyte in fish.

The study of morphological figures and morphometrical data observed in lung buds from ETAS, cultured in Agar and OPEP medium, with or without SCAPEP, at different concentrations, in order to detect its influence on the morphogenesis, has shown that: OPEP (5:0) supports gradual morphogenesis for a limited time; OPEP + SCAPEP (5:2) supports gradual morphogenesis for a prolonged time; OPEP+SCAPEP (5:5) supports fast and deep morphogenesis for a short time; OPEP+SCAPEP (5:7,5) causes early inhibition of morphogenesis and survival; SCAPEP (5:0) stops instantly morphogenesis and survival.

The factors governing the pattern formation process in the early morphogenesis of a marine colonial hydroid, Dynamena pumila, have been studied. Two different types of morphogenesis have been distinguished. Morphogenesis of the first type goes on via changes in cell shape and cell axis orientation, while morphogenesis of the second type is based upon the active coordinated cell movements associated with cell rearrangements. It was shown that morphogenesis of both types can be considered as cascades in which any event is a consequence of the previous one. The spatial structure of each developmental stage contains information about the direction and the initial conditions of further morphogenesis. So, an "epigenetic program" of morphogenesis gradually originates in the course of development and provides the stable reproduction of spatial structures. It is reasonable to consider the activity of epigenetic factors guiding Dynamena morphogenesis (geometry/topology of an embryo, heterogeneity of an embryo spatial structure, configuration of the field of mechanical stresses of the embryo surface) as "morphomechanical programming" of morphogenesis.

By pathologic findings, the author studied the infection of purified hepatitis virus by oral administration, and compared it with those by the other infectious routes. The results are briefly summarized as follow:1) The establishment of infection by oral administration of the purified virus could be well proved by observing the pathologic findings. Compared with those caused by oral administration of the crude virus, the purified virus caused severer pathologic findings and mecroid or necrotic parts were widely observed without exception.2) The infection by the purified virus through the other routes was also well established, though it was often weaker than that through the mouth.3) Compared with that of the crued virus, the inoculation of the purified virus produced more marked pathologic findings which made the determination of infection easier.These results suggests that the application of the purified virus made the infectious exeriment a step of advance.

Treated with anti-microtubule drugs—amiprophos-methyl, the effect of cortical microtubules disassembly on the ultrastructure of cucumber young leaf cells was investigated by transmission electron microscope in this paper. The results were as follows:treated with 15×10 ˉ6 APM, the growth of cucumber seedling was inhibited. Following the time, the ultrastructure showed that intercellular spaces became larger, vacuoles were separated, chloroplasts deformed and moved to the central section of the cell, and starch grains in chloroplasts almost disappeared.These results suggested that the changes of cell ultrastructure might be produced by the microtubules disassembly .
