    To establish methods for quantitative determination of ginseng saponins, ginsenoside Rg1, Re, Rb1 and polysaccarides and compare the qualities of Tongrentang Red Ginseng and Korean Red Ginseng.Macroreticular resin-colorimetric method was developed to determine ginseng saponins and a new HPLC method with gradient eluents was established for determination of ginsenoside Rg1, Re, Rb1. For ginseng polysaccharides, phenol-oil of vitriol colorimetric method was developed and some factors were also optimized.The content of ginseng saponins in Tongrentang Red Ginseng was not lower than that of Korean Red Ginseng. Ginsenoside Rg1 and Rb1 in Tongrentang Red Ginseng were higher than those in Korean Red Ginseng, while Ginsenoside Re was slightly lower than that of Korean Red Ginseng. However, the amount of Ginseng Polysaccharides in Tongrentang Red Ginseng was greater than those in Korean Red Ginseng.The contents of ginseng saponins and ginsenoside Rg1, Re, Rb1 in Tongrentang Red Ginseng were not lower than that in Korean Red Ginseng. The methods for determination of ginsenosides and ginseng polysaccharides were quite accurate and reliable to the quality control of Ginseng.

    Panax ginseng is one of the most medicinally used herbal medicines in the world. Wild ginseng is widely accepted to be more active than cultivated ginseng in chemoprevention. However, little has actually been reported on the differences between wild ginseng and cultivated ginseng.To identify wild ginseng-specific genes, we used suppressive subtraction hybridization.We report that one of the clones isolated in this screen was the GAPDH(glyceraldehyde 3-phosphate dehydrogenase) gene (designated pGAPDH-w). DNA BLAST sequence analysis revealed that this pGAPDH-w gene contained novel sequences of 94 bp. RT-PCR results showed that the expression of the pGAPDH-w gene was significantly up-regulated in the wild ginseng as compared with the cultivated ginseng.The pGAPDH-w gene may be one of the important markers of wild ginseng.

    F. Ginseng (Panax ginseng) is planted in the forest to enhance the natural ginseng resources, which have an immense medicinal and economic value. The morphology of the cultivated plants becomes similar to that of wild growing ginseng (W. Ginseng) over the years. So far, there have been no studies highlighting the physiological or functional changes in F. Ginseng and its wild counterparts. In the present study, we used proteomic technologies (2DE and iTRAQ) coupled to mass spectrometry to compare W. Ginseng and F. Ginseng at various growth stages. Hierarchical cluster analysis based on protein abundance revealed that the protein expression profile of 25-year-old F. Ginseng was more like W. Ginseng than less 20-year-old F. Ginseng. We identified 192 differentially expressed protein spots in F. Ginseng. These protein spots increased with increase in growth years of F. Ginseng and were associated with proteins involved in energy metabolism, ginsenosides biosynthesis, and stress response. The mRNA, physiological, and metabolic analysis showed that the external morphology, protein expression profile, and ginsenoside synthesis ability of the F. Ginseng increased just like that of W. Ginseng with the increase in age. Our study represents the first characterization of the proteome of F. Ginseng during development and provides new insights into the metabolism and accumulation of ginsenosides.

    Abstract: Hemodialysis deteriorates oxidative stress. Vitamin E is an antioxidant whose regeneration is provided for by vitamin C. The authors tested the effects of a vitamin E‐modified membrane (E), nonmodified cellulose membrane (O), and vitamin C infusion (500 mg, C) into the arterial blood line during dialysis on parameters of oxidative stress. In a short‐term study, 24 patients were subjected to a single dialysis session with E, O, E with C, and O with C protocols. In a long‐term study (12 weeks), 20 patients were randomized into groups with C and without C on each dialysis, and both groups had dialysis using O, E, and again O membrane for 4 weeks each. In the short‐term study, thiobarbituric acid reacting substances (TBARS) in plasma rose after dialysis (p < 0.02) with O, and no changes were observed in the other 3 protocols. In the long‐term study, predialysis TBARS declined when using E both in the groups with C (p < 0.02) and without C (p < 0.05). A switch over to O resulted in TBARS returning to baseline levels. The E membrane prevented an increase in lipid peroxidation during single dialysis, and long‐term use of the E membrane also resulted in a decrease in the predialysis lipid peroxidation level. The antioxidant capacity of the E membrane was not enhanced by vitamin C infusion. High doses of vitamin C administered during dialysis using a nonmodified cellulose membrane prevented an increase in lipid peroxidation, most probably due to the enhanced rate of endogenous vitamin E regeneration.

    The antioxidant effects of vitamins C and E on cryptorchidism-induced oxidative stress were investigated in male Sprague-Dawley rats. Forty rats (200-250 g) were randomly divided in a blinded fashion into five groups (n = 8). Group 1 was sham operated and treated with vehicle (corn-oil, 10 mL/kg). Groups 2, 3, 4, and 5 were rendered unilaterally cryptorchid and treated with vehicle (10 mL/kg), vitamin E solution (75 mg/kg), vitamin C solution (1.25 g/kg), and combination of vitamin E (75 mg/kg) and vitamin C (1.25 g/kg) solutions, respectively. Germ cell count, superoxide dismutase (SOD), total protein (TP), and testicular weight (TW) were lower, but malondialdhyde (MDA) was higher in the cryptorchid rats than the sham-operated rats. When administered separately, vitamins C and E increased germ cell count, SOD, TP, and TW but did not reduce MDA in the cryptorchid rats when compared to the vehicle-treated cryptorchid rats. However, there was no significant difference in these parameters between vehicle-treated and combined vitamins C- and E-treated rats. This suggests that vitamins E and C alleviate the germ cell loss and oxidative stress in cryptorchidism when administered separately but not when combined in rats.

    Objective: To research differences between Ginseng protein from Panax ginseng of different origins. Methods: The extraction methods of ginseng protein and the experiment conditions of membrane hybridization were established,and peptide arrays method was applied to hybrid with ginseng protein. Results: There were significant differences in Ginseng protein from P. ginseng of different origins,in which the difference was significant between high latitude regions( XK、ML) and low latitude regions( CB、JA、XS),and the difference was not obvious between the same latitude regions( XK and ML or CB、JA、XS). Conclusion: The polypeptide array technology can be used to research the ginseng protein of P. ginseng of different origins and different growth conditions. Ginseng protein from different regions showed obvious difference. In addition,ginseng protein expression characteristic map can be used for the identification of P. ginseng.

    The patterns of ginseng saponins in the commercial ginseng tea samples and each parts of ginseng plant were investigated by quantitative thin-layer chromatography. The quality of those sample teas were also evaluated. (1) White ginseng contained about times of Ra(o) than did other parts of ginseng. (2) Lateral roots, peelings and buds of ginseng were rich in , , c, which constituted about 50% of total saponin. (3) The ratio of Rb.c to Rg(f) in the leaves and stems of ginseng plant was 0.64 : 1. (White ginseng, 2 : 1 ; buds, 3 : 1 ; flower, 3.2 : 1 ; peelings, 5.8 : 1 ; lateral ginseng, 7 : 1) The relative content of Rg(f) in the white ginseng was about 3 times as much as the lateral ginseng. (4) The ratios of panaxadiol to panaxatriol in 13 kinds of commercial ginseng teas were in the range of .

    Objective: To compare the effect of sun-dried ginseng, red ginseng and mild property ginseng on Qi deficiency of rats. Methods: Swimming strain method and Caloric restriction method were applied to copy Qi deficiency syndrome model of rats, which were given sun-dried ginseng, red ginseng and mild property ginseng suspension gavage respectively; the effect on qi deficiency rats was compared. Results: Sun-dried ginseng, red ginseng and mild property ginseng can increase rats fatigue endurance swimming, reduce the content of cAMP and cAMP/cGMP ratio, elevate the content of cGMP, as well as reduce the activity of LDH. Conclusions: Sun-dried ginseng, red ginseng and mild property ginseng and provide therapeutic effect of qi deficiency rats, without significant difference between the high dose group. un-dried ginseng, mild property ginseng are better than red ginseng, just in values.
