    Abstract:
    Diastereomeric 2-methoxycyclopentyl analogues of a natural quorum sensing signaling molecule from Pseudomonas aeruginosa were synthesized and screened in pigment production assays with P. aeruginosa and Serratia strain ATCC39006.

    EDTA is highly toxic for Pseudomonas stutzeri. The bactericidal effect is not significantly attenuated by treatment of the cells at 0 degrees C rather than 25 degrees C, nor by osmotic stabilisation of the treated cells. The few surviving cells are not genetically resistant, but resistance to EDTA (and to some cationic antibiotics) can be induced by growing organisms in magnesium-limited media. Under these conditions, there is increased production of a protein of molecular mass about 20 kD. Comparable results have been reported for Pseudomonas aeruginosa. The mode of action of EDTA on these pseudomonads is discussed.

    Suspensions obtained from five Pseudomonas aeruginosa pyocinogenic strains showed inhibitory and variable activity against bacterial strains belonging to the Nocardiaceae, Micrococcaceae, Neisseriaceae, Streptococcaceae, Vibrionaceae, Enterobacteriaceae, and Pseudomonadaceae families. Under special conditions, the same pyocinogenic P. aeruginosa strain can be affected by it own suspensions. These pyocinlike particles could be considered as a regulatory factor acting on the rate and size of the population growth.

    Many bacteria use cell-cell communication to monitor their population density, synchronize their behaviour and socially interact. This communication results in a coordinated gene regulation and is generally called quorum sensing. In gram-negative bacteria, the most common quorum signal molecules are acylated homoserine lactones (AHLs), although other low-molecular-mass signalling molecules have been described such as Autoinducer-2 (AI-2). The phenotypes that are regulated in Serratia species by means of AHLs are remarkably diverse and of profound biological and ecological significance, and often interconnected with other global regulators. Furthermore, AHL- and AI-2-mediated systems (less profoundly studied) are continuously being discovered and explored in Serratia spp., many having interesting twists on the basic theme. Therefore, this review will highlight the current known quorum sensing systems in Serratia spp., including the important nosocomial pathogen Serratia marcescens.

    Pseudomonas fluorescens 410PR grows on 4-nitrobenzoate but does not metabolize 4-nitrotoluene. The TOL pWW0 delta pm plasmid converts 4-nitrotoluene into 4-nitrobenzoate through its upper pathway, but it does not metabolize 4-nitrobenzoate. P. fluorescens 410PR(pWW0 delta pm) transconjugants were isolated and found to be able to grow on 4-nitrotoluene. This phenotype was stable after growth for at least 300 generations without any selective pressure. P. fluorescens 410PR(pWW0 delta pm) converted 4-nitrotoluene into 4-nitrobenzoate via 4-nitrobenzylalcohol and 4-nitrobenzaldehyde. 4-Nitrobenzoate was metabolized via 4-hydroxylaminobenzoate and finally yielded NH4+ and 3,4-dihydroxybenzoate, which was mineralized.

    Abstract A fluorescent pseudomonad inciting brown angular leaf spots on iron wood ( Parotia persica ) in Mazandaran forest was isolated and identified as a pathovar of Pseudomonas syringae. Strains assimilated adonitol and L‐tartrate but not lactate or D‐tartrate as carbon sources for growth. The electrophoretic profiles of cell proteins of strains isolated from iron wood were very similar but differred markedly from protein profile of P. syringae pv. syringae.

    Monoclonal antibody MAI-6 directed against Pseudomonas aeruginosa strain PAO1 outer membrane lipoprotein H2 also interacted with a protein having an identical molecular weight from 50 of 52 strains of P. aeruginosa tested. This antibody interacted with the P. fluorescens group of the family Pseudomonadaceae and with Azotobacter vinelandii but not with several other species of gram-negative bacteria. A colony blotting assay performed with this antibody allowed rapid detection of the P. fluorescens-A. vinelandii group.

    The effect of a commonly used biocide, 1,2-benzisothiazolin-3-one (BIT) on ppGpp accumulation in the pathogen, Pseudomonas aeruginosa PAO1, and an environmental isolate, Ps. fluorescens, was examined. It is concluded that BIT is able to induce a stringent response in Ps. aeruginosa and Ps. fluorescens, determined by the rapid accumulation of ppGpp following addition of BIT to exponentially-growing cells. Western blot analysis of whole-cell extracts with anti-RelA antibody demonstrated that both species contain a RelA homologue. This is the first report of a RelA-like protein in pseudomonads.

    N-Acyl-l-homoserine-lactone-producing Serratia species are frequently encountered in spoiling foods of vegetable and protein origin. The role of quorum sensing in the food spoiling properties of these bacteria is currently being investigated. A set of luxR luxI homologous genes encoding a putative quorum sensor was identified in the N-(3-oxo-hexanoyl)-l-homoserine lactone (3-oxo-C6-HSL)-producing Serratia proteamaculans strain B5a. The 3-oxo-C6-HSL synthase SprI showed 79 % similarity with EsaI from Pantoea stewartii and the putative regulatory protein SprR was 86 % similar to the SpnR of Serratia marcescens. Proteome analysis suggested that the presence of at least 39 intracellular proteins was affected by the 3-oxo-C6-HSL-based quorum sensing system. The lipB-encoded secretion system was identified as one target gene of the quorum sensing system. LipB was required for the production of extracellular lipolytic and proteolytic activities, thus rendering the production of food-deterioration-relevant exoenzymes indirectly under the control of quorum sensing. Strain B5a caused quorum-sensing-controlled spoilage of milk. Furthermore, chitinolytic activity was controlled by quorum sensing. This control appeared to be direct and not mediated via LipB. The data presented here demonstrate that quorum-sensing-controlled exoenzymic activities affect food quality.
