    Abstract:
Phthalide-3-carboxylic acids decarboxylate readily in the presence of imines, the product depending on the solvent used. In dimethyl sulfoxide, or in the absence of solvent, at 130, the product is the 3-alkyl 3-hydroxyisoindolone or its dehydration product, but in acetic anhydride at 130 the product is a mixture of diastereoisomeric 3-acetylarninoalkylphthalides. The reactions can be applied to the synthesis of natural products such as the isoindoloisoquinoline and phthalideisoquinoline alkaloids.

The reaction of 1-acetyl-2-phenylhydrazine with ethoxymethylenemalononitrile yielded [(4-cyano-1-phenylpyrazol-3-yl)aminomethylene]propanedinitrile. Hydrolysis followed by annulation with methyl isocyanate provided a synthetic route to 2-phenylpyrazolo[3,4-d]pyrimidines.

β-Hydroxy acids react with various aldehydes in the presence of acidic materials to give spiro lactones in good yields. For example, 2-isopropyl-5-methyl-1,3-dioxaspiro[5,5]undecan-4-one was obtained in 90% yield from the reaction of 2-(1'-hydroxycyclohexyl)propionic acid and isobutyraldehyde.

Biocatalysis offers mild reaction conditions, an environmentally attractive catalyst–solvent system, high activities, and chemo-, regio-, and stereoselectivities, while the use of enzymes generally circumvents the need for functional group activation and avoids protection/deprotection steps required in traditional organic syntheses. This review, using β-lactam antibiotics as an example, discusses recent advances in biocatalysis research towards the goal of ‘green’ methodologies for the manufacture of (fine) chemicals and the emulation of a cell's enzymatic cascade processes.

Birch-type reductions of 12-methoxypodocarpa-8,11,13-trien-19-01 (2) have been investigated and the products have been converted into the 13-oxo derivatives (7) and (30) which have potential use as chiral intermediates for synthesis. The C14 ketone (9) which is also of potential value, has been prepared from the reduced derivative, methyl 12-oxopodocarp-13-en-19-oate (21).
