Objective: An HPLC method for the simultaneous determination of Chlorogenic acid and Caffeic acid in Flos Lonicerae was proposed based on rapid ultrasonic-assisted extraction using the non-ionic surfactant TritonX-100 as the extractant. Methods: The separation was performed on Phenomenex C18( 250 mm ×4.6 mm,5 μm) column with detection wavelength of 326 nm. The mobile phase consisted of methanol-0. 6% acetic acid solution( 25 ∶ 75,V /V) with the flow rate of 0. 80 mL /min,and the column temperature was set at 35℃. Result: The calibration curves for Chlorogenic acid and Caffeic acid were liner between 0. 00076 ~ 190. 00 μg /mL( r = 0. 99995),0. 000864 ~ 216. 00 μg /mL( r =0. 99966) respectively. The limit of detection( LOD) were 0. 029 ng /mL,0. 030 ng /mL,respectively. The recoveries of the two compounds range were 95. 13% ~ 100. 24%. Conclusion: The method was simple,rapid,convenient,sensitive,and environment-friendly,and thus can provide new and scientific means for assaying the contents of Chlorogenic acid and Caffeic acid in Flos Lonicerae.

Objective:Comparing Se-enriched Salvia miltiorrhiza and Salvia miltiorrhiza content of effective components.Methods:A high performance liquid chromatographic method determination.Results:In Se-enriched Salvia miltiorrhiza fat-soluble components TanshinoneⅡA content of about 2.7times higher than in Salvia miltiorrhiza;In Se-enriched Salvia miltiorrhiza water-soluble components Danshensu content of about 3 times higher than in Salvia miltiorrhiza.Conclusion:Se-enriched cultivating Salvia miltiorrhiza can improve the content of active ingredients in Salvia miltiorrhiza.

Objective:To prepare and establish a method for the determination of baicalin liposomes.Methods:Baicalin liposome was prepared by reverse evaporating method,the content of baicalin in baicalin lipsomes was determined by HPLC.Results:The mean diameter and entrapment efficiency of baicalin lipsomes were 350nm and 60.1%.The linear range of baicalin was 0.29~1.45μg(r=0.9997) and the average recovery rate was 100.1%,RSD=0.2%(n=6).Conclusion:The preparation of baicalin lipsomes is simple and the content determination is simple,reliable and reproducible,which can be used in the quality control of baicalin lipsomes.

A gradient reversed-phase HPLC method was developed for the simultaneous separation and determination of rosmarinic acid, lithospermic acid B, caffeic acid, and other phenolic compounds. This method showed a satisfactory result. The limits of detection for these phenolic compounds were ≤0.05 mg/L. The developed method was applied to the determination of phenolic compounds in the dried roots of Salvia miltiorrhiza (Danshen) and the transformed cells of S. miltiorrhiza. In the transformed cells, rosmarinic acid was the predominant phenolic compound. Small amounts of lithospermic acid B and caffeic acid were also detected. In the dried roots, in contrast, lithospermic acid B was the main phenolic compound and only a very small amount of rosmarinic acid was found. In addition, danshensu and some unidentified phenolic compounds were also detected from the dried roots. Objective: To establish the quality standard for the Jing Salvia miltiorrhiza Bge and the Jiu Salvia miltiorrhiza Bge.Methods: The Jing Salvia miltiorrhiza Bge and the Jiu Salvia miltiorrhiza Bge was identified by TLC.Results: The methods could identify the characteristic spots of the Jing Salvia miltiorrhiza Bge and the Jiu Salvia miltiorrhiza Bge.Conclusion: This method is simple and special in operation,the results are stable with a good repeatality.These can be well applied to control the quality of the Jing Salvia miltiorrhiza Bge and the Jiu Salvia miltiorrhiza Bge.

Simultaneous determination of caffeic acid and baicalin in Shuanghuanglian injection by HPLC-DAD-ECD
Objective:A simultaneous determination of caffeic acid and baicalin in Shuanghuanglian injection by HPLC- DAD- ECD has been established.Method:The samples were separated on Zorbax SB-C_(18)column (150 mm×4.6mm,5.0μm)by a gradient elution program with methanol and 0.6% acetic acid [0 min,A-B(25: 75);5 min,A-B(28:72);10 min,A-B(37:63);13 min,A-B(55:45)] as mobile phase at a flow rate of 0.8 mL·min~(-1).The detections were done at 279nm,0.6 V and 25℃.The standard curve method was used for the quantitative detection of caffeic acid and baicalin.Results:The linear ranges of caffeic acid and baicalin were 0.35 -33.5μg·mL~(-1)(r =0.9997)and 1.30-1.25×10~3μg·mL~(-1)(r =0.9994),respectively.The recoveries were 108% (RSD=1.2%,n=5)and 99.7% (RSD=1.6%,n=5),respectively.Conclusion:The method is rapid, accurate and reproducible.It can be used for quality control of Shuanghuanglian injection.

OBJECTIVE To establish a quality standard of Xiaozhi capsule.METHODS The identification of Radix et Rhizoma Rhei,Flos sophorae were carried out by TLC.The content of baicalin in Xiaozhi capsule was determined by HPLC. RESULTS Radix et Rhizoma Rhei,Flos Sophorae could be idetifid by TLC.A good linearity of baicalin was obtained in the range of 0.1002~1.6032 μg(r=0.999 8) and the average recovery of baicalin was 99.3%,RSD was 0.98%.CONCLUSION This method is simple,sensitive and accurate,and has good reproducibility.It can be used for the quality control fo Xiaozhi Capsule.

Objective To probe into the anti-endotoxin effects of baicalin isolated from Radix Scutellariae. Methods A 0.5% aqueous solution of baicalin was prepared which was extracted and isolated from Radix Scutellariae.The in vitro antiendotoxin effect of baicalin was quantitatively determined with the limulus test after a mixture of endotoxin and baicalin had been incubated in a 37 ℃water bath for 1 h.The in vitro inhibitory of baicalin on the endotoxin-induced fever was measured in rabbits.The LPS-induced deaths in mice pretreated and not pretreated with baicalin were compared.Effects of baicalin on the LPS induced excessive increase in the serum contents of tumor necrosis factor-α(TNF-α) and nitric oxide(NO) were studied in mice. Results 0.833 mg· mL~(-1) of baicalin was shown to result in a degradation of endotoxin from 4 EU to 0.546 EU,the destruction percentage being 87.45%.5 mL·kg~(-1)of 0.5% baicalin solution injected intravenously per rabbit gave rise to a striking abatemant of the endotoxin-induced fever.The LPS-induced death rate in mice pretreated with baicalin(10%) was significantly lower than that in those without baicalin pretreatment(70%).The excessive increase in the serum TNF-α and NO induced by LPS in mice was brought down significantly by baicalin in a dose-dependent manner. Conclusion Baicalin isolated from Radix Scutellariae was shown to be provided with anti-endotoxin effects.
