Use of the antiandrogen cyproterone acetate in the treatment of hirsutism.--25 women with hirsutism or hypertrichosis faciei were subject to a "reverse sequential therapy" with cyproterone acetate (Hammerstein's regimen) for 1 to 3 years. In 8 patients, a limited, in 14 patients a satisfactory and in one woman a good result could be recorded. Cycle irregularities and subjective side effects were within a tolerable range. Studies of the hair pattern in 15 patients revealed a higher telogen rate as well as increased diameters of the hair stalks, compared with controls, in the temporal region of the head. These alterations disappeared in the course of the treatment. In the parietal region of the head no significant differences of the hair pattern were found in comparison with the controls and during application of the steroids.

Cyproterone acetate の投与による睾丸の造精機能および内分泌機能の変化を雄ラットで検討する目的で, 成熟雄ラットに cyproterone acetate を10mg/kg/day連日2週間又は4週間投与し, 投与終了時に血漿中LH, FSH, testosterone 値を測定し, 電気刺激射精法によつて得られた精液中精子数を算定した. さらに摘出睾丸潅流法により睾丸の progesterone, 17α-hydroxyprogesterone および testosterone 分泌能を測定し, 睾丸間質の組織学的所見と併せて対照群と cyproterone acetate 投与群と比較検討した. Cyproterone acetate 投与後の睾丸重量は, 対照群と比べ有意差はなく, 組織学的には, 精細管精子細胞の変性が認められたが, 睾丸間質細胞数には対照群と差はなかつた. 射精液中の精子数は対照群で742±200×104 (mean±SEM) に対し, cyproterone acetate 投与群で399±89×104と減少したが両者間に有意差はなかつた. 血中LH, FSH値は本剤投与により有意に上昇し (p<0.001), 逆に testosterone 値は有意に減少した (p<0.05). 睾丸潅流法による testosterone 分泌能は, 対照群の3.39±0.67ng/minに対し, 投与2週後で1.55±0.67ng/minと減少し, 投与4週後には0.91±0.21ng/minと有意に抑制された (p<0.001). また cyproterone acetate 投与により, 睾丸の progesterone 分泌能は対照群と比べ有意差はなかつたが, 17α-hydroxyprogesterone 分泌能は有意に増加した (p<0.05). この事実から cyproterone acetate が睾丸の17, 20 desmolase もしくは 17ketosteroid reductase 活性を抑制する可能性が示唆された.

(1) The antiandrogenic steroid cyproterone acetate exerted a strongly negative nitrogen balance in 8 young male volunteers, the degree of which correlated with the dose administered. In a male patient of 77 years with cancer of the prostate, this catabolic effect proved to be much weaker. In 8 female patients with hirsutism, administration of cyproterone acetate resulted only then in a negative nitrogen balance when the food was deficient in calories and protein. Continuous treatment with cyproterone acetate in female patients is not feasible because of strongly progestational side-effects. These side effects can be avoided by combining cyproterone acetate with ethinyl estradiol as an ‘inverted sequential scheme’, according to Hammerstein and Cupceancy [2]. In most female patients with hirsutism, good or satisfactory results can be obtained by a continuous cyproterone acetate therapy or by the combination treatment of Hammerstein and CUPCEANCY [2].

Abstract The theoretical assumptions to understand the elution mechanism of the discontinuous solution fractionation and complete elution method could be confirmed for the system poly(vinyl acetate)/butyl acetate/cyclohexane. In this case the process of complete elution, however, can only be carried out with an unfavorable high consumption of time and material. The complete elution method with all its advantages can be preserved, if the process is carried out as multistage process with a limited number of discontinuous steps. By means of such a multistage procedure improved separation results were achieved compared with the conventional single stage procedure. Furthermore some results are given which show a decrease of the separation effect of solution fractionation due to procedural limitations.

Abstract Several polypropene samples, synthesized with different catalytic systems based on TiCl4 supported on activated MgCl2 and different electron donors, were studied using both preparative and analytical temperature-rising elution fractionation (TREF). The experimental results show that each variation in the catalytic system is clearly reflected in the nature of the corresponding polymer, which can be distinguished by both preparative temperature rising elution fractionation and analytical temperature rising elution fractionation dissolution profiles.

The administration of cyproterone acetate can be effective in the treatment of ‘idiopathic’ hirsutism. In 5 similarly affected patients treated with ethinyloestradiol and lynoestrenol (Minilyn) the rate of hair growth was reduced in only two, even though the serum total testosterone was reduced in each instance.

Twenty-four girls with sexual precocity who had been treated with cyproterone acetate were followed for periods from 1 to 8 1/2 years after the discontinuation of therapy. It was found that their further pubertal development and the resumption or appearance of menstruation followed a natural course within the wide limits of physiological variability. It is concluded that puberty resumes its natural course following cessation of treatment with cyproterone acetate, and it is assumed that the reproductive ability of these patients will not be affected.
