The teachings of Sad Dharma become an important basis that can be used by educators in creating learning strategies that are able to direct students to apply the theories of religious education that they get. The values of Sad Dharma's teachings are also very relevant to be applied at the level of education, both from Kindergarten (TK) and even college. The implementation of the Sad Dharma-based strategy is also part of the preservation of Balinese Hindu culture such as the dharma gita and dharam discourse which are rarely touched by the younger generation. What Sad Dharma are 6 dharma goals that must be carried out by Hindus indirectly through the implementation of Sad Dharma-based Hindu Religious Education strategies, educators are able to deliver their students to achieve these goals. The purpose of this study was to find out about the Sad Dharma-based Hindu education strategy. The research method is qualitative using secondary data sources, data collection techniques in this study use study documentation plus library techniques and when the data has been obtained and compiled in the form of scientific work, the author will perform data analysis. Overall, this sad dharma-based Hindu education strategy can provide maximum results in the quality of student education because its implementation is not only found in theory but also in action in implementing its teachings in line with the concept of learning that is currently being promoted by the government.

The Mahāyana Mahāparinirvāṇasūtra （MPM）, a middle period Mahāyāna sūtra, introduces Abhirati, the Buddha-land of Akṣobhya, as the place in which the Dharma protectors who defend the pure bhikṣu, the keeper of precepts, from the violence of the corupt monks who will appear in the latter days of Buddhism are reborn. In addition, in his previous life, the Buddha Śākyamuni was a Dharma protector who risked his life to protect the pure bhikṣu, and through the merits of that time he attained the Vajra body. On the other hand, the Akṣobhyavyūha （AV）, an early Mahāyāna sūtra, does not refer to Abhirati as the land of the Dharma protectors. However, it refers to the downfall of Dharma. MPM attributes this to “the appearance of bhikṣus who do not keep the precepts,” while the AV explains that the reason for this is that “people no longer have the desire to listen to the Dharma, and the Dharma preachers no longer teach the Dharma.”

To screen the learning and memory mutant from N-ethyl-N-nitrosourea (ENU) mutagenic zebrafish F1, and to get the new model animal to study the mechanism of learning and memory.Zebrafish mutant was screened by inhibitory avoidance behavioral test and identified by the expression of gene c-fos with qRT-PCR.We isolated a zebrafish mutant related to learning and memory, fgt. In this fgt zebrafish mutant long-term memory was much lower than that in wild-type when tested at 24 h after training. The 24 h long-term memory in about half of fgt mutant F2 (13/30) were significantly lower than those in wild-type, and the others relatively normal. Compared with the expression in wild-type fishes, the expression of immediate-early genes (IEGs) c-fos in half of fgt mutant F2 (13/30) after exploring in a novel environment increased distinctly from the basal control levels statistically, and the others relatively normal, which were in accordance with the behavioral results.The zebrafish mutant fgt is a dominant mutant with defect in long-term memory.

In Tanluanʼs Commentary on the Treatise on the Pure Land浄土論註, we find a passage quoted from Sengzhaoʼs Commentary on the Vimalakīrtinirdeśa 注維摩経. It is applied to the background of the concept of ʻthe two kinds of dharma-body.’ This concept consists of the following five ideas: 1. Buddhas and Bodhisattvas; 2. Buddhaʼs Dharma-nature and Bodhisattvas’ expedient means for saving beings; 3. The two kinds of Dharma-Body （Buddhaʼs Dharma-Body as dharma-nature and Bodhisattvas’ Dharma-Body as expedient means） originate in each other; 4. The inexpressible aspect of Buddhaʼs reality and expressible aspect of Bodhisattvas’ expedients interact with each other; 5. The two kinds of Dharma-Body are to be identified as ‘Dharma.’

Οι καρδιαγγειακές παθήσεις αποτελούν στην εποχή μας μία από τις πρώτες αιτίες θανάτου στον ανεπτυγμένο κόσμο. Επίσης, πολύ σημαντική είναι η συμβολή της αγγειογένεσης στην ανάπτυξη των καρκινικών όγκων. Καθίσταται λοιπόν αναγκαία η συμβολή της βασικής έρευνας ώστε να μελετηθούν καλύτερα οι μηχανισμοί των παθήσεων αυτών αλλά και να ανακαλυφθούν νέα φάρμακα που σχετίζονται με την αγγειογένεση. Τα τελευταία χρόνια σημαντικό ρόλο παίζει προς αυτές τις κατευθύνσεις το ζωϊκό πρότυπο μοντέλο zebrafish. Το zebrafish (Danio rerio) χρησιμοποιείται ευρέως ως ζωικό μοντέλο για την κατανόηση της παθοφυσιολογίας των καρδιαγγειακών παθήσεων. Στην παρούσα διατριβή παρουσιάζεται ο καρδιακός φαινότυπος υποτροφικού κόλπου, myh6-/-, σε ενήλικα ψάρια, τα οποία φέρουν μετάλλαξη στη βαριά αλυσίδα κολπικής μυοσίνης του zebrafish. Οι ομόζυγοι φορείς επιβιώνουν μέχρι την ενηλικίωση και είναι γόνιμοι. Στα ενήλικα μεταλλαγμένα ψάρια, ο κόλπος παραμένει υποπλαστικός και εμφανίζει εναπόθεση ελαστίνης, ενώ οι κοιλίες παρουσιάζουν αυξημένο μέγεθος. Στα θηλαστικά, η υπερτροφία είναι ο συνηθέστερος μηχανισμός που οδηγεί σε καρδιομεγαλία. Χρησιμοποιώντας την ανοσοϊστοχημεία και συνεστιακή μικροσκοπία σάρωσης για να μετρήσουμε το μέγεθος, την πυκνότητα και τον πολλαπλασιασμό των καρδιομυοκυττάρων, αποδεικνύεται ότι η μεγέθυνση της κοιλίας myh6-/- οφείλεται κατά κύριο λόγο στην υπερπλασία. Εντούτοις, εντοπίσαμε παρόμοια μεταγραφικά προφίλ στην απόκριση υπερτροφίας θηλαστικών μέσω RT-PCR των υπερπλαστικών κοιλιών. Περαιτέρω, δείχνουμε την ενεργοποίηση της οδού του στρες-ΕΔ με ανάλυση ανοσοαποτυπώματος κατά Western. Συμπερασματικά, μπορούμε να υποθέσουμε, με βάση το μοντέλο μας, ότι μονοπάτια μοριακής σηματοδότησης που συνδέονται με την υπερτροφία στα θηλαστικά, σε συνδυασμό με την ενεργοποίηση του στρες-ΕΔ, καταλήγουν σε υπερπλασία στο zebrafish. Επιπλέον, αυτή είναι η πρώτη φορά που αναφέρθηκε η εναπόθεση ελαστίνης στον κόλπο. Παράλληλα, μελετήθηκαν μικρά μόρια που αναστέλλουν την αγγειογένεση και είναι ελπιδοφόρα υποψήφια φάρμακα για τον καρκίνο, τις αμφιβληστροειδοπάθειες και τον εκφυλισμό της ωχράς κηλίδας που σχετίζεται με την ηλικία. Η in vivo φαινοτυπική διαλογή σε zebrafish (Danio rerio) αναδεικνύεται ως μια ισχυρή μεθοδολογία για την ταυτοποίηση και βελτιστοποίηση νέων ενώσεων με φαρμακολογική δράση. Το zebrafish παρέχει αρκετά πλεονεκτήματα για τις in vivo φαινοτυπικές παρατηρήσεις ειδικά για την αγγειογένεση, καθώς αναπτύσσεται ταχέως και δεν βασίζεται σε λειτουργικό καρδιαγγειακό σύστημα για να επιβιώσει για αρκετές ημέρες κατά τη διάρκεια της ανάπτυξης. Σε αυτή τη μελέτη, χρησιμοποιούμε μια διαγονιδιακή σειρά που επιτρέπει τη μη επεμβατική παρακολούθηση της αγγειογένεσης σε κυτταρικό επίπεδο. Η αναστολή της αγγειογένεσης μπορεί να παρατηρηθεί κάτω από ένα στερεοσκόπιο φθορισμού και να ποσοτικοποιηθεί. Εκμεταλλευόμενοι τις δυνάτοτητες του zebrafish, χρησιμοποιήσαμε μια σειρά από 60 καινοτόμες ενώσεις που σχεδιάστηκαν με βάση την αναστολή του υποδοχέα VEGFR2. Στα πειράματα αυτά μπορεί να να υπολογιστεί η συγκέντρωση στην οποία προκαλείται τοξικότητα και να καταδειχθούν πιθανές παρενέργειες των υπό εξέταση ουσιών. Με αυτόν τον τρόπο αποκαλύφθηκαν 6 αντιστρεπτοί αναστολείς της αγγειογένεσης. Τέλος, στα πλαίσια της παρούσας διδακτορικής διατριβής επιχειρήθηκε να ανιχνευθούν γονίδια τα οποία παίζουν ρόλο στη μορφογένεση των καρδιακών βαλβίδων και να αποδειχθεί η σχέση τους μέσω στοχευμένης μεταλλαξιγένεσης με τη βοήθεια του συστήματος CRISPR-Cas και να δημιουργηθεί μια διαγονιδιακή σειρά zebrafish που να αποκρίνεται στις αλλαγές της διατμητικής τάσης. Συμπερασματικά, από την παρούσα διατριβή προκύπτει η μεγάλη δυνατότητα του zebrafish για ανασύσταση/αναδιαμόρφωση της καρδιάς του σε παθολογικές καταστάσεις και αποδεικνύεται η ικανότητά του να χρησιμοποιείται ως ιδανικό μοντέλο για τη μελέτη των καρδιαγγειακών παθήσεων.

Education directs a person to make a better life. Through education can change a person into a character who has noble character, has sympathy and empathy for all creatures. The recent reality is that education only directs the achievement of intellectual intelligence but forgets emotional, spiritual and skill intelligence. It is necessary to re-develop holistic education by implementing learning models or methods in the educational process. The objectives to be achieved in the preparation of this research are to know about holistic education and the implementation of the Dasa Dharma learning method in holistic education. The method used is descriptive qualitative research. The results of this study can provide an overview of holistic education. Holistic education is education as a whole which includes intellectual, emotional, physical, social, aesthetic and spiritual potential. Various kinds of learning methods that can be used in holistic education. One of them is the dasa dharma method as the implementation of holistic education. The dasa dharma method includes dharma carita, dharma lila, dharma gita, dharma kriya, dharma shanti, dharma wacana, dharma yatra, dharma tula, dharma sadhana, and dharma brata.
