Abstract:
Affinity-based fluorescence sensing has been one of the key enabling technologies in biomolecular sensing, used for detection of proteins, DNAs, toxins, bacteria, etc, and remains one of the most sensitive, specific, robust, and widely used diagnostics methodology [1-4]. In absence of high-performance integrated optical filters, miniaturization of a fluorescence sensing system in CMOS has relied on time-resolved techniques with synchronized sources or externally grown optical filters and/or collimators. This paper presents a nanophotonic-electronic co-design approach towards fully-integrated fluorescence biosensor with on-chip copper-interconnect based nanoplasmonic filters. The filters demonstrate a measured extinction ratio of greater than 51dB in the excitation/emission bands for a class of quantum-dot based fluorescence tags. Integrated with these filters, the sensor platform is a correlated double sampling architecture which achieves femtowatt photon sensitivity. Detection sensitivity of 47 zeptomoles of quantum-dots was experimentally demonstrated, making the chip a low-cost, fully integrated, high-performance, and fully scalable biosensor for point-of-care applications.Keywords:

Love waves, a variety of surface acoustic waves (SAWs), can be used to detect very small biological surface interactions and so have a wide range of potential applications. To demonstrate the practicality of a Love wave SAW biosensor, we fabricated a 155-MHz Love wave SAW biosensor and compared it with a commercial surface Plasmon resonance (SPR) using glycerol-water solution with known densities and viscosities to calibrate the response signals of the biosensors. And the mass per unit area of anti-mouse IgG bound with protein G onto the sensitive layer of the biosensor was calculated on the basis of the calibration result. The sensitivity of the Love wave SAW biosensor was the same as or greater than that of the SPR biosensor. Furthermore, the Love wave SAW biosensor was capable of measuring a much wider range of viscosities than the SPR biosensor. Although the operating principle of the Love wave SAW biosensor is completely different from that of the SPR biosensor, the subtle changes in the viscoelastic properties of the biological layer that accompany biological binding reactions on the sensitive layer can be monitored and measured in the same ways as with the SPR biosensor.

The objective behind the development of miniaturization or small specimen test technology is to reduce the cost and quantity of material involved during the characterization of the material. The idea of the development of miniaturization took attention when the nuclear industry starts developing as these materials are very costly and it is not economically feasible to waste large amount of these materials for the sole purpose of testing. The second factor which promotes the miniaturization is that the working of machine is not affected while at the same time its material is being tested. At present, the idea of miniaturization is being applied to other materials also. The miniaturization of standards for metals has been done successfully in the past. For composites, not much work has been done. In the chapter, the specimen size effects on tensile properties of glass fiber composite have been identified by varying the length and width simultaneously and have established a relationship between the ASTM standard specimen and the small size specimen.
