To study the effect of saponins of asparagus on apoptosis and the variations of Caspase-8, 9, 3 activity in the process of asparagus induced apoptosis in HepG-2. Cell was administrated with different concentrations of asparagus at different time, the apoptosis rate was determined by FCM with AV-PI staining, the morphology was observed by electron microscopy, Caspase-8, 9, 3 activities was measured by kit. Experiments showed that saponins of asparagus inhibited cell growth. At 72 h, the apoptosis rate increased with concentrations markedly. Apoptotic morphology was observed .The activities of Caspase-8, 9, 3 increased with concentrations markedly, which had significant difference compared with negative control group. The activity of Caspase-8 is high at 24 h and the activities of Caspase-9 and Caspase-3 is high at 48 h. Saponins of asparagus can inhibit the growth of HepG2, and the mechanism may be related to regulating Caspase-8, 9 activity, which subsequently actives Caspase-3.

To explore the relationship between apoptosis of liver cells and Caspase-3 in endotoxemia(ET) and antagonistic effect of Cation A(CA) in rats.Seventy-two adult healthy SD rats were randomly divided into 3 groups,including control group,ET group and CA group.Hepatic tissues in three groups were collected for tests at 3rd,4th,8th and 12th hour after treatment.Apoptosis of liver cells were detected by flow cytometry,and Caspase-3 expression was detected by immunohistochemical staining.The results showed that the percentage of apoptosis of liver cells in ET group were significantly higher than that of control and CA group.The percentage of hepatocyte apoptosis of ET group was in an upward trend,however,which was upward trend in the CA group at the 3rd,4th,8th hours time points,and went downward trend at the 12th hour.Caspase-3 expression in ET group was significantly higher than that of CA group.These results suggested that apoptosis and Caspase-3 protein expression in liver cell was positive correlation in endotoxemia,and CA have a significantly antagonistic effect on inflammatory injure through down-regulation of the expression of Caspase-3 pathway in liver,inhibiting liver cell apoptosis

Objective To study on whether the apoptosis existed and the change of gene expression related to apoptosis by investigation of apoptosis of the cerebral contusion and expression of Bcl-2/Bax and caspase-3 related to apoptosis in human.Method We researched that apoptosis and gene expression of Bcl-2/Bax and caspase-3in brain tissues by FCM and immunohistochemical.Results Analysis showed that apoptosis of neuro-cell was related to patient's injury serious level and injury time(P0 05),but was not related to prognosis.The positive expression of Bcl-2 was related to prognosis(P0 05),but was not related to sex?age?GCS and injury time.The caspase-3 was highly correlated with the apoptosis,which showed the expression of caspase-3 was an indicator of apoptosis degree.Conclusions Apoptosis occurred in human brain after actue injury which fulfilled a function in pathological transformation of brain tissues. The expression of Bcl-2 was related to prognosis. The expression of caspase-3 could reflect the levels of apoptosis.

Asparagus is a perennial herb rich in multiple nutrients; however, the utilization of asparagus is lower on account of one-third part of the asparagus often be discarded as a by-product at harvest in order to standardize the length for market. In this study, asparagus spears were divided into tip (top of the spear, 5 cm), mid (intermediate of the spear, 5 cm), and base (rest of the spear, 5 cm) parts, and the mid part and base part were cut into as 2–3 cm length. As following, the three parts of asparagus spears were processed into tea with green tea processing craft, respectively. Electronic nose and GC-MS analyses were used to investigate the aroma components of three asparagus teas. A total of 59 components were detected, among which there were 37, 43, and 50 components in the tip, mid, and base asparagus tea, respectively. According to the electronic nose analysis and GC-MS analysis, we speculate aldehydes, alkanes, and ketone were the main aroma components of asparagus tea. And the different aroma components among the three asparagus teas might be caused by the different spear parts of asparagus. The sensory evaluation score of the mid asparagus tea was 88.54 ± 0.33, which was highest among the three teas. The results indicated that mid asparagus tea might be more popular with consumers. Practical applications Asparagus (Asparagus officinalis L.) were divided into tip, mid, and base parts, and every part was processed into tea respectively. The process optimization and aroma analysis could improve the utilization rate of asparagus and also provides a basis for clarifying the specific volatile of asparagus tea.
