Expression of Leptin-receptor in human dental pulp stem cells

Objective: To isolate,culture and identify human dental pulp stem cells(hDPSCs) and investigate the expression of Leptin-receptor in hDPSCs.Methods:hDPSCs were cultured with tissue-explants and enzyme-digestion techniques.Immunochemistry and immunofluorescence were used to detect the expression of cytokines in hDPSCs.The differentiation potential was evaluated by adipogenic and osteogenic induction assay.The expression of Leptin-receptor in hDPSCs was detected with strept avidin-biotin peroxidase complex(SABC).Results:Vimentin and CD146 were expressed in hDPSCs,hDPSCs had the potentials to differentiate into adipocytes and odontoblasts.Positive expression of Leptin-receptor in hDPSCs indicated hDPSCs were target cells for Leptin action.Conclusion:hDPSCs can be cultured by both tissue-explant and enzyme-digest techniques,carrying characters of undifferentiated mesenchyme cells.The expression of CD146 and Leptin-receptor are detected in hDPSCs.
