Effects of Different Media on the Saponin Contents in the Calli of Panax notoginseng Pollens

[Objective] The study aimed to discuss the content changing of total saponin during the callus culture of Panax notoginseng pollens on different media.[Method] With MS medium as basic medium,different hormones(IAA,NAA,KT,6-BA,GA,2,4-D and HL) and nutrient matters(sucrose and agar) were added to prepare 5 different kinds of media.The effects of different media on the saponin contents in the calli of P.notoginseng pollens were studied.[Result] During the extraction of total saponin,the extraction content of saponin was higher when the temperature was set at 70 ℃ and 30% ethanol was used.The contents of total saponin were different in the calli of P.notoginseng pollens cultured on different media,among the total saponin contents on 5 kinds of media,the highest was 3.45%,the lowest was 0.02% and the difference between them was 172.5 times.[Conclusion] The addition of 2,4-D in medium may promote the synthesis of saponin in the calli of P.notoginseng pollens,so the content of total saponin was increased.
