Pseudomonas aeruginosa biofilm formation: a proteomic study.

OBJECTIVE To discover different proteins among Pseudomonas aeruginosa(PAE) mucoid PDO300,wide-type P.aeruginosa PAEO1,PAEO1 lasI rhlI mutants and PAEO1 lasR rhlR mutants strains.METHODS The protein mass spectra of the tropina of mucoid PDO300,wide-type PAEO1,PAEO1 lasI rhlI and PAEO1 lasR rhlR mutants strains were detected by CM10 protein chip on surface enhanced laser desorption/ionization/time of flight/mass spectrometry(SELDI),then analyzed by Biomarker Wizard software.RESULTS Sixteen different proteins were found between mucoid PDO300 and wide-type PAEO1(P0.01).Ten proteins with relative molecular mass(Mr) 4074,3817,8923,8148,3332,4521,7621,11 594,7831 and 13 751 were up-regulated while six proteins with Mr 6199,6899,12 379,6115,6763 and 8576 were down-regulated in the tropina of PDO300.Comparing the tropina of wide-type PAEO1 and PAEO1 lasI rhlI mutants strains,there was one protein down-regulated in PAEO1 lasI rhlI mutants strain(P0.05).It′s Mr 6910.There were four different proteins between wide-type PAEO1 and PAEO1 lasR rhlR mutants strains(P0.05).Two proteins with Mr 8924 and 8 790 were up-regulated while two proteins with Mr 6735 and 6986 were down-regulated in the tropina of PAEO1 lasR rhlR mutants strain.CONCLUSIONS In this study,we have taken advantage of the techniques of SELDI to detect variation in protein expression among different P.aeruginosa strains.These different proteins may contribute to the phenotypic differences or have relation to quorum sensing(QS) system.
