转录因子Ets-1对P21WAF1/CIP1启动子的影响

AIM: To observe the effects of transcription factor Ets-1 on P21WAF1/CIP1 promoter. METHODS: Transient transfection and luciferase assay were used to analyze the effects of Ets-1 on the activity of the P21WAF1/CIP1 promoter construction with lucifersase reporter gene. RESULTS: Ets-1 increased P21WAFI/CIP1 transcription. Dominant negative Ets-1 failed to activate P21WAF1/CIP1 transcription. Ets-1 interacted selectively with the (-1350)^GGAA^(-1347) Ets element in the P21WAF1/CIP1 promoter. Mutation of this element reduced basal and Ets-1-inducible P21WAF1/CIP1 promoter-dependent expression. In contrast, the (-1577)^GGAT^(-1574) motif mediated basal but not Ets-1 activation of the P21WAF1/CIP1 promoter. CONCLUSION: Ets-1 regulates P21WAF1/CIP1 transcription by binding to (-1350)^GGAA^(-1347) element in the P21WAF1/CIP1 promoter.
