Propagación in vitro de explantes de Teca obtenidos a partir de semillas

The teak plants can be cultivated starting from seeds, which are very important to maintain wide genetics base. The thick and hard pericarp blocks the germination and a considerable part of the fresh seeds remain latent during the first year. Therefore the application of the tissue culture techniques constitutes a valuable tool for the obtaining of teak plants starting from seeds. The objective of the present work was to germinate in vitro teak seeds, to establish the conditions and culture medium for its multiplication and to evaluate the possibility of the ex vitro rooting. The seeds were placed in a culture medium composed by the salts MS and 6-Bencilaminopurina (6-BAP). The germinated plants were subcultivated in different culture medium for the multiplication with several concentrations of 6-BAP and of Kinetin (KIN). The time of subculture, the handling type to be carried out with the explants and the physical state of the culture medium also, was evaluated, and in the rooting phase the feasibility of the ex vitro rooting was studied. The employment of the 6-BAP and the KIN to reason of 2.0 and 0.5 mg.l -1 respectively allowed the formation of a bigger number of buds and a bigger multiplication coefficient, in additionit was obtained that the best time of subculture was every 28 days using culture medium in semisolid state, and the handling that contributed to the biggest multiplication coefficient consisted on the separation of the buds and cut of the explants bigger than 2.5cm in to two parts.
