Time-resolved fluoroimmunoassay of #beta#_(2)-microglobulin and its preliminary appliation

The two-site time-resolved fluoroimmunoassay (TRFIA) was established in this study for the detection of (#beta#_(2)-m). The tested speciment or #beta#_(2)-m reference standard was added to the micro-titer plates coated with sheep anti rabbit IgG antibody, and diluted rabbit anti-#beta#_(2)-m antibody and Eu~(3+)-#beta#_(2)-m reaction solution were added. After incubation at 25℃ for 1.5 hours and washing for 4 times, the enhancement solution was then added. The fluorescence testing was performed by Auto DELFIA-1235. The intra-and inter-assay CV of this TRFIA were 2.25% and 2.91% respectively. The recovery rate was 101.06% and the sensitivity was 0.06mg/L, with a range of testing from 0.06mg/L to 12mg/L. There was no cross reaction with albumin and myohemoglobin,and hemolysis showed no any influence on this method. The result of preliminary application was consisten with those of radioimmunoassay.
