The invention discloses a nucleic acid-ribosome combination system construction method and application thereof. The nucleic acid-ribosome combination system construction method comprises the following steps: adding 12 mul of ribosome wheat germ extraction solution to a 500mul- centrifuging tubelet which is treated by 0.1% DEPC (diethylpyrocarbonate), then sequentially adding 1mul of RNAsin (40U/mul, Takara), 2mul of solution containing 100mM of KAC, 4mM of Mg(AC)2, 20mM of Hepes and 20mM of DTT (Dithiothreitol), 1mul of 1mM/each 20 amino acid standard mixed liquor (Promega), 2.5mul of 10mM of ATP, 1mul of 10mM of GTP, 0.5mul of 2mM of cycloheximide, 1.5mul of 8mM of phosphocreatine, 1mug of TMV-RNA and 0.5mul of creatine phosphate kinase, adding DEPC treating water till the volume is 25mul, centrifuging, putting at the temperature of 23-26 DEG C for 14-16 minutes, and then putting at the temperature of 0-5 DEG C to obtain the nucleic acid-ribosome combination system. The nucleic acid-ribosome combination system construction method is applied to research of drug screening. A nucleic acid-ribosome combination system constructed by the method can be applied to research of viral nucleic acid-ribosome combination suppressive drug screening.
