Studies on genetic stability of sweetpotato germplasm maintained in National In Vitro Genebank

Three mark methods,morphological characterization,the esterase(EST)isozyme electrophoretogram and random amplified polymorphism DNA(RAPD)molecular marker were used to analyse genetic stability of sweetpotato germplasm maintained In Vitro for 8 years at the National Sweetpotato In Vitro Genebank.The results showed that most agronomic characters and the EST isozyme zymogram of sweetpotato germplasm stored in vitro for a long term were basically the same as the controls preserved in field.With molecular marker RAPD,majority varieties had the same amplified product,and few difference was detected in individual varieties.That means long term in vitro storage may affect the genetic stability of individual sweetpotato germplasm,and this affection could be detected by RAPD.
