Propofol precondioning alleviating myocardiac ischemia/reperfusion injury on rats is related to inhibition of mitochondria permeability transition

Aim To observe the relationship between propofol precondioning alleviating myocardiac ischemia/reperfusion injury and mitochondria permeability transition.Methods Myocardiac regional ischemia/reperfusion model was established via clamping left anterior descending coronary artery(LADCA) in vivo.Adult male SD rats were randomly assigned into four groups:Sham group,subjected to constant perfusion 3 h,ischemia/reperfusion group(IR),underwent ischemia 60 min and reperfusion 120 min,DMSO group and propofol precondioning(PP) group,respectively infused DMSO and propofol for 20 min before clamping LADCA at same rate.Results Undergoing 120 min reperfusion,PP group apoptosis index was statistically lower than that of IR group(24.35±2.35 vs 33.38±3.47,P0.05).Observed with electron microscope,mitochondria ultrastructure in IR group was destroyed totally while mitochondria membrane potential losed statistically,compared with Sham group(0.25±0.06 vs 0.91±0.24,P0.05).PP group preserved relatively intact mitochondria ultrastructure and maintained membrane potential compared whit IR group(0.67±0.13,P0.05).Otherwise,DMSO group had no protective effects.Conclusions Propofol precondioning can alleviate myocardiac ischemia/reperfusion injury on rats and it is related to inhibition of mitochondria permeability transition.
