The invention belongs to the field of genetic engineering, and discloses a fusion protein mGM-CSF-X10-beta hCGCTP37 peptide, and a preparation method and application thereof. A fusion expression preparation technical platform of the laboratory is used for constructing an engineering bacterium of fusion expression mGM-CSF-X10-beta hCGCTP37 peptide in an Escherichia coli cell. Ion exchange chromatography is used for separating a target protein. The correct expression of the fusion protein and the immunogenicity of an epitope in the fusion protein are verified and detected by in-vitro Western bloting. In-vivo animal experiments expect that a DC vaccine obtained after DC is sensitized by the in-vitro impact of the fusion protein can be transmitted back into the body to endow APC with high antigen loading and handling capability, improve the immune response of organisms to tumor cells and fulfill the aims of identifying and killing the tumor cells, and the fusion protein has broad clinical application prospect in anti-tumor treatment.
