Impact of the myocardial ischemia reperfusion on apoptosis in the boundary area, and its significance:Experimental study

2013
Objective To study impact of the myocardial ischemia reperfusion on apoptosis in the boundary area, and its significance. Methods The myocardial ischemia/reperfusion model in rabbit was build up;relations of dynamic expression of boundary caspase-3 mRNA and bcl-2 mRNA,and myocardial cell apoptosis were assessed by the technique of semi-quality RT-PCR and conventional HE.Results From 4 h after ischemia reperfusion injury caspase-3 mRNA and apoptosis started to lift,at the 12 h reached the high peak.A significantly positive correlation was found between the number of apoptotic myocytes and logarithmic value of caspase-3 mRNA expression(r=0.9286,P0.001);while ischemia reperfusion injury at the 12 h bcl-2 mRNA started to be activated, then kept on to maintain the low level appearance, untill 48 h then the apoptosis and bcl-2 mRNA expression were significantly negatively correlated(r=-0.9648,P0.001).Conclusion Capase-3 and bcl-2 dysequilibrium is an important cause inducing boundary area cell apoptosis which will influence ventricular reconstitution.
