The Effect of Physiological and Biochemical Characteristics of Wheat Seedling with Agrobacterium tumefaciens Soaking Roots

Using resplit-plot design,wheat genotypes Xinong 979,Xinong 889,and Zhengmai 9023 as main plot,root length 4,7 and 10 cm as split plot,and Agrobacterium tumefaciens concentration 0,0.5,1.0 and 1.5 OD as split-split plot,the biochemical characteristics of wheat seedlings were measured after soaking wheat seedlings root with Agrobacterium tumefaciens.It was found that the growth rate,root activity,POD activity and SOD activity of Xinong889 were higher than Xinong979 and Zhengmai 9023;with the increasing of root length by Agrobacterium tumefaciens soaking,the root growth rate,seedling root length,seedling fresh mass,root activity,and POD activity increased as well,while the seedling height showed a downward trend,molality of solute MDA increased when root length increased in the range of 4-7 cm,and in the range of 7-10 cm the molality of solute MDA did not change significantly;compared with the control,when Agrobacterium tumefaciens concentration was at 0.5 OD,wheat root growth rate,seedling fresh mass,and root activity increased slightly.With the increasing of concentration of Agrobacterium tumefaciens,wheat root growth rate,seedling fresh mass,and root activity significantly decreased.While molality of solute MDA levels had been increasing,POD activity and SOD activity increased and then showed a downward trend.According to the effect of soaking root treatment with Agrobacterium tumefaciens to the main physiological and biochemical characteristics of wheat seedling,the Agrobacterium tumefaciens bacterial concentration of 0.5 OD and root length of 10 cm are appropriate for soaking root transformation,and appropriate genotype for soaking root is Xinong889.
