Application on Thiazolyl Blue(MTT) Plaque Forming Assay to Determine Virus Titer in Virology

Objective:To evaluate the application of thiazolyl blue(MTT) plaque forming assay to determine virus titer in virology.Methods: To perform a plaque forming assay,influenza virus and RSV were inoculated onto MDCD and Hep-2 cell monolayers,a single layer nutrient agar was added to cover cells and cultured for 3-4 days,MTT was used instead of neutral red to stain the cells for counting virus plaque.Results:(1)Compared to the neutral red plaque forming assay,MTT plaque forming assay was easier and faster;(2)Accuracy rating of the MTT plaque forming assay was the same as the neutral red plaque forming assay;(3)The false plaque forming of the MTT plaque forming assay was lower than the neutral red plaque forming assay.Conclusion: The MTT plaque forming assay could be used to replace the neutral red plaque forming assay,and it is more rapid and accurate.
