Study on Expression Levels of STC-1,HIF-1α and Ca~(2+) Contents in Renal Carcinoma Cell line Affected by Stanniocalcin-1

Objective:To investigate whether STC-1,HIF-1α involved in the pathogenesis of renal cell carcinoma through adjusting the content of Ca2+ in cells.Methods:Human renal cell carcinoma cells(GRC-1) were treated with different concentrations of exogenous human STC-1 protein,Cell growth of GRC-1 was detected by assay of MTT.Ex-pression of STC-1,HIF-1α was tested by polymerase chain reaction.Fluorescence probe was used to label GRC-1 and intracellular Ca2+ were observed with the fluorescence detector device.Results:Exogenous STC-1 protein can stimulate the proliferation of GRC-1 cells.Compared with the control group,its role in promoting the proliferation rate was gradually decreased with the increase of the concentration of exogenous STC-1 protein(P 0.05).Exogenous STC-1 protein can reduce the expression of STC-1,HIF-1α and Ca2+ levels in GRC-1 cells and this effect was positively correlated with STC-1 concentration(P 0.05).Conclusions:STC-1,HIF-1α may involv in the pathogenesis of renal carcinoma by mutual inhibition and regulation of intracellular Ca2+ levels.
