HPLC Determination of Aloenin in Leaves and Preparations of Aloe arborescens Mill.

Amethod for quantitative determination of aloenin by microcolumn reversed-phase HPLC with UV detection (306 nm) using a KAH-6-80-4 column (2 × 80 mm, Separon-C18, 7 μm), mobile phase MeCN–aqueous HOAc (1%) (25:75), elution rate 100 μL/min, and eluent volume 2000 μL was developed. Validation showed that the method was characterized satisfactory metrological parameters. The limit of quantitation for aloenin was 115 mg/mL. The accuracy at an aloenin content level within 80 – 120% was 96.54 – 103.28%. The method was used for analysis of Aloe arborescens Mill. leaves, ex tempore obtained juice, and some medicines (Aloe liquid extract for subcutaneous injection, Aloe juice). Statistical processing indicated that the relative error of average results for the aloenin assay did not exceed 5% with a confidence probability of 95%.
