Model Establishment of Alcohol Liver Injury in Mouse and Effects of the Extract from Schisandra on the Injury

Objective To establish the mouse model of alcohol liver injury whose mechanism is similar to human's and to investigate the effects of the alcohol extract from Schisandra Sphenanthera Rdhd.et.wils.(Schisandra) on alcohol liver injury in mice.Methods Complex liquid food with alcohol was made,and the ICR mice were divided into five groups including normal control group,experimental group Ⅰ,experimental group Ⅱ,experimental group Ⅲ and experimental group Ⅳ.the mouse model of alcohol liver injury were made by complex liquid food and LPS.Experimental group Ⅲ and experimental group Ⅳ were fed by liquid alcohol food supplemented with 0.5% and 1.5% alcohol extract of Schisandra respectively.After 8 weeks,experimental groups were injected with 1mg/Kg lipopolysaccharide(LPS) by caudal vein,after 6 hours,all mice were killed,and the content of ALT,AST,GGT in blood were determined and the pathologic changes of liver were observed.Results The mouse model of alcohol liver injury was successfully established by complex alcohol liquid food and LPS,the activities of ALT were increased significantly from 16.3±5.2U/L to 52.6±18.7U/L,while the activities of ALT in blood after Schisandra treatment were decreased significantly(P0.05).Local degeneration of hepatic cell were observed,after injecting LPS,the nucleus were largened and dyed strongly.Conclusion The mouse model of alcohol liver injury was successfully established by complex alcohol liquid food and LPS,and the alcohol extract of Schisandra can reduce alcohol liver injury.
