Genetic Relationship of Wormwood Capillary (Artemisia capillaris Thunb.) In Perak as Revealed by RAPD

Random amplified polymorphic DNA (RAPD) technique has been used to detect polymorphism of Wormwood (Artemisia capillaris Thunb.) in the State of Perak, Malaysia. The genomic DNA was extracted from the leaves of A. capillaris using Sarkosyl method which successfully produced a good purity and quality of DNA. Ten (OPA 04, OPA 09, OPA 16, OPA 17, OPA 18, OPG 03, OPG 05, OPG 09, OPG 13 and 391) out of 57 oligonucleotide screened primers were selected to amplify DNA of five populations of A. capillaris. A total of 368 RAPD fragments with 75% of them being polymorphic ranging from 250-3000 bp were scored from the populations. Genetic distance among the populations ranged from 0.17 to 0.51 and the similarity index ranged from 0.556 to 0.842.
