Expressions of TGF-β1 in the murine model of chronic obstructive pulmonary disease and the intervention of tetrandrine

Objective To study the expression of TGF-β1 in the murine model of chronic obstructive pulmonary disease(COPD)and the intervention of tetrandrine.Methods 10 rats were assigned to the normal group and 40 to the model of COPD group that was established by an intratracheal infusion of small amount of endotoxin and smoke.Then the latter group was randomly divided into COPD control,Qi-deficiency,Yin-deficiency,and blood stasis group and the latter three were administered with tetrandrine.Expressions of TGF-β1 were detected in bronchi and lung tissues.Results TGF-β1 in the epithelium of the bronchi was mildly expressed in the normal control group,highly expressed in the COPD control group,and moderately expressed in the three groups with tetrandrine intervention.The relative content of TGF-β1 was higher in the model of COPD group than in the normal control group(0.457 ± 0.047 vs.0.213 ± 0.034,P 0.05=.The TGF-β1 level was 0.250 ± 0.019 in the Qi-deficiency group,0.270 ± 0.028 in the Yin-deficiency group,and 0.242 ± 0.034 in the blood stasis group after the intervention of tetrandrine,all of which were lower than that in the COPD control group(P 0.01=.There were no differences among the three intervention groups.Conclusion Tetrandrine can reduce the thickening of the smooth muscles of the bronchi,inhibit the expression of TGF-β1,and suppress the formation of remodeling of the airway due to COPD.
