Expression of retrovirus mediated HSV1-TK gene in human renal cancer cell line GRC-1

Objective To investigate the expression and significance of HSV1 TK gene in human renal cancer GRC 1 cells . Methods A retrovirus expression vector G1Na TK was constructed with pgk tk cDNA and a retroviral vector G1Na.The G1Na TK vector was transfer into renal cancer GRC 1 cells with lipofectin reagent and the HSV TK vector producer cell line GRC 1/TK was isolated in 1mg/ml neomycin medium. Results The HSV TK vector producer cell line GRC 1/TK was obtained which confered a chemosensitivity to ACV. Conclusions HSV1 TK gene might be used in gene therapy of human renal cancer GRC 1 cells.
