ABSTRACTIntroduction: Levodopa induced dyskinesia (LID) is a common motor complication affecting almost 95% of patients after 15 years of levodopa therapy. Dyskinesia in Parkinson`s disease (PD) can be functionally and socially disabling. Amantadine ER/ADS-5102/Gocovri™ is the first and only drug to get FDA approval in August 2017 for treatment of dyskinesias in patients with PD receiving levodopa with or without concomitant dopaminergic medication.Areas covered: This review summarizes the pharmacodynamics, safety and efficacy of Amantadine ER/ADS 5102 in the treatment of LID.Expert opinion: LID is a challenging problem in Parkinson’s disease. Although Amantadine IR is effective in treating LID, it is associated with a higher risk of CNS related side effects including sleep disturbances.
