EFFECT OF THEABROWNIN IN PU-ERH TEA ON THE HORMONE-SENSITIVE LIPASE ACTIVITY AND mRNA EXPRESSION IN RAT

2010
Objective To explore the theabrownin(TB) in Pu-erh tea on hormone-sensitive lipase(HSL) activity and mRNA expression in rat liver and epididymis adipose tissue.Method Ninety SD rats were randomly divided into normal control group(Group Ⅰ),high-fat model group(Group Ⅱ) and high-fat+TB group(Group Ⅲ ).Ten rats were selected from each group and killed at D 15,D 30 and D 45 of the experiment respectively.TG、TC、LDL-C and HDL-C in serum and HSL activity and mRNA expression level in liver and epididymis tissue were detected.Results(1) In D 30 and D 45 of the experiment,TG、TC and LDL-C in serum in Group Ⅱ were higher than that in Group I(P0.05,P0.01),but HDL-C was lower than that in Group I(P0.05,P0.01),while TG、TC and LDL-C in serum in Group Ⅲ were lower than those in Group Ⅱ(P0.05,P0.01),but HDL-Chigher than that in Group Ⅱ(P0.05,P0.01).(2) In D 30 of the experiment.HSL activity in epididymis adipose tissue in Group Ⅲ was higher than that in Group I(P0.05).while in D 45,HSL activity in liver in Group Ⅲ was higher than that in Group I(P0.05).HSL activity in epididymis adipose tissue in Group Ⅱ and Group Ⅲ were higher than that in Group I(P0.05,P0.01).(3) In D 30 of the experiment,HSL mRNA expression of epididymis adipose tissue in Group Ⅱ and Ⅲ was respectively higher than that in Group I(P0.05,P0.01),and the difference between the latter two was also significant(P0.01).In D 45 of the experiment,HSL mRNA expression in liver tissue in Group Ⅲ was significantly higher than that in Group Ⅰ(P0.05),while mRNA expression in epididymis adipose tissue in Group Ⅱ and Ⅲ was higher than that in GroupⅠ(P0.05,P0.01),and the difference between Group Ⅱ and Ⅲ was extremely significant (P0.01).Conclusion Pu-erh tea TB has the hypolipidemic effect to some extent and significantly enhances HSL mRNA expression in liver and epididymis adipose tissue, thereby increase HSL activity in rat.It is one of the mechanisms of TB’s hypolipidemic effect.
