Expression and significance of HMGB1 and NF-κ B in hydatidiform mole and choriocarcinom

Objective:To explore the change of expression of high mobility group protein(HMGB1)and nuclear factor-κBp65(NF-κBp65)in normal pregnancy,hydatidiform mole and choriocarcinom,further reveal the biological characteristics of the cells of placental site trophoblastic tumor. Methods:Specimen from abortion,cesarean section,hydatidiform mole and choriocarcinoma patients were collected in Weifang Hospital of Traditional Chinese Medicine and Weifang People′s Hospital from 2010 to 2013.The expression of HMGB1 and NF-κBp65 in early pregnancy,the placenta in late pregnancy and hydatidiform mole and choriocarcinoma were detected with immunohistochemical method. Results:1.HMGB1 positive cells were detected in early pregnant,late pregnancy,hydatidiform mole and choriocarcinoma which were localized in the cytoplasm and nucleus. mainly distributes in trophoblasts and vascular endothelial cells and interstitial cells. Compared with the early pregnant and late pregnancy,the intensity of the HMGB1 positive cells is remarkably strengthened in hydatidiform and choriocarcinoma,and there was the statistical significance(P0.001). 2.Immunohistochemistry result showed that the NF-κ B p65 positive granules mainly locate in the cytoplasm,mainly distributes in trophoblasts and vascular endothelial cells and interstitial cells. Compared with the early pregnant and late pregnancy,the intensity of the NF-κ B p65 positive cells is remarkably strengthened in hydatidiform and choriocarcinoma,the difference has the statistical significance(P0.001). 3.The average optical density of the HMGB1 and NF-κ Bp65 in the hydatidiform mole is a positive correlation(r=0.7499,P=0.000). The HMGB1 and NF-κ Bp65 in choriocarcinoma is a positive correlation(r=0.7338,P=0.000). Conclusion:1.It suggests that the high expression of the HMGB1 and the NF-κ Bp65 was closely related to hydatidiform and choriocarcinoma. 2.The expression of the HMGB1 and NF-κ Bp65 in hydatidiform and choriocarcinoma have the same trend. Linear correlation analysis is positive. It suggests that the HMGB1 high expression may strengthen the expression of NF-κ Bp65,which is related to the growth of tumor cell.
