Influence of somatostatin analogue on human hepatocellular cancer cell line MHCC97-H

AIM: To study the influence of somatostatin analogue octreotide on human hepatocellular cancer cell line MHCC97 -H and its mechanisms in vivo. METHODS: Twenty-seven nude mice,of which MHCC97-H tumor mass was planted in liver, were divided into 3 groups randomly: negative control group, low dose interfered group, large dose interfered group,and raised at similar condition for 35 d the survival time , the weight at the death point were racorded and the pathologicail changes of livers and lungs were changed; the expression of matrix metalloproteinases-2(MMP-2)in hepatocellular carcinoma was detected by immunohistochemical technique. RESULTS: Compared with the animal models of negative control group, the animal models of interfered groups were heavier in mean weight at death point and lower in metastasis rate (P0.05); while there was no statistical difference between the 2 interfered groups (P0.05); the expression of MMP-2 in hepatocellular carcinoma of interfered groups was gradually increased than that in control group(P 0.05); while there was no statistical difference between interfered groups(P0.05). CONCLUSION: Octreotide can improve the living quality of animal models and inhibit the metastasis of human hepatocellular cancer cell line MHCC97-H in vivo,which might be related to the down-regulation of the expression of MMP-2.
