Experimental Study on the Mouse Model of Paraquat Poisoning In duced Pulmonary Fibrosis by Intraperitoneal Injection of Paraquat

Objective: To establish a stable mouse model of paraquat poisoning induced pulmonary fibrosis by a single intraperitoneal injection of paraquat.Methods: 60 healthy female C57BL/6J mice were randomly divided in to six groups according to the paraquat dosage: the saline control group,and the 30,40,50,60 and 80 mg/kg paraquat group,each group of 10 mice.The mice were sacrificed on day 28 for pathological examination(HE stain ing).The LD50 was calculated and the sections after HE staining were evaluated for the formation of lung fibrosis by using Ashcroft scoring system.Results: The LD50 of intraperitoneal paraquat of 28 days was 55.1923 mg/kg.All the paraquat dosage could induce some degrees of pulmonary fibrosis.The severity and early mortality in creased with the dosage.Conclution: We successfully established a mouse model of pulmonary fibrosis by single intraperitoneal injection of paraquat.The optimal paraquat dosage is 40 or 50 mg/kg.
