A flow cytometer excitation light source system and a correction method are disclosed. The system comprises a light source, an adjusting device, a light splitting and detecting device, a shaping and focusing device and a feedback control device, wherein the adjusting device is connected to the feedback control device and used for receiving excitation light emitted from the light source, adjusting the propagation direction of the excitation light according to control signals from the feedback control device and then allowing the light to emerge; the light splitting and detecting device is connected to the feedback control device and used for splitting light emergent from the adjusting device into two beams, detecting light intensity and light spot positions of one of the two beams, transmitting signals including information of the light intensity and the light spot positions to the feedback control device and allowing the other beam to emerge; the shaping and focusing device is used for shaping and focusing the light emergent from the light splitting and detecting device and allowing the light to emerge to an object; and the feedback control device is used for receiving signals including the information of the light intensity and the light spot positions with the signals being transmitted by the light splitting and detecting device, calculating an adjusting quantity and a light intensity correction according to the signals, and transmitting control signals containing the adjusting quantity to the adjusting device. The system and the method are advantaged by high alignment precision.
