Analysis of pear fruit acid /low-acid trait by SSR marker

The inheritance of the fruit acid trait as titratable acid of mature fruit was analyzed using the F1 segregation population(total of 118 seedlings) derived from the cross Bayuehong(Pyrus communis L.) × Dangshansuli(P.× bretshneider Rehd.).Meanwhile,225 pairs of SSR primers originated from pear and apple genome were screened by bulked segregant analysis(BSA) and selective genotyping(SG).Of those primer pairs,only primer KU10 produce a polymorphic band linked to acid trait,the genetic distance of the molecular marker is 12.15 cM,and the quantitative trait loci(QTL) analysis of fruit acid/low-acid trait was mapped on second linkage group(LG2) of the pear genetic map.The comprehensive analysis of molecular marker and phenotype revealed that the fruit acid trait is dominant to low-acid trait and is affected by both additive gene effect and environmental factors,and it is also affected by non-additive gene effect.
