Effect of qindan capsule on expression of vascular adventitial fibroblasts remodling in hypertensive rats

Objective To investigate the effects of qindan capsule(QC)on vascular adventitial fibroblasts remodling in spontaneously hypertensive rats(SHRs).Methods Thirty-two SHRs of 40 weeks old were divided into the model(group C),high dose QC(group D),low dose QC(group E),losartan(group F).Sixteen Wistar rats were taken as the normal controls(group A)and QC group(group B).Blood pressure was recorded every three weeks.After 12 weeks,the thoracic aorta in groups of D,E and F was taken for observing the morphological changes and nuclear density by HE staining and laser scanning confocal microscope,and detecting the mRNA expression of transforming growth factor β1(TGF-β1)in thoracic aorta by immunohistochemical staining and real-time quantitative fluorescent PCR.Results Compared with group A,QC could reduce the blood pressure of SHRs,increase the ratio of ID/mesolamella/adventitia of endangium and the expression of TGF-β1.Conclusion QC could not only reduce blood pressure of SHRs,but also suppress and even reverse adventitial vascular remodeling.The mechanism may be related to the down-regulation of TGF-β1 mRNA expression in thoracic aortic wall and inhibition of myofibroblasts transformation.
