The Determination of Growth,Physiological,Biochemical Parameters and the Development of SCAR Marker of the New Strain ZC of Gracilariopsis lemaneiformis(Rhodophyta)

Quantitative traits of growth rate,physiological and biochemical parameters including total protein content,pigment contents and the rates of photosynthetic oxygen evolution and respiration oxygen consumption were determined and measured among four strains of Gracilariopsis lemaneiformis,one of which was collected in Zhanshan Bay,Qingdao,the other two strains were 981 and 07-2cultivars,and the new strain named ZC.The aim of the study was to provide assistant evidence for cultivation application of this strain.The linear growth rate of the new strain was up to 9.89 mm/d and increased by 16.6%、51.7% more than that of 981 and 07-2respectively,which illustrated that growth rate of the new strain was higher than the existing cultivars.Its phycoerythrin content was significantly lower than 07-2and had no significant differences with 981.However,its chlorophyll a content was markedly lower than two cultivars.Total protein content(7.21mg/g)of the new strain was the highest among four strains,which demonstrated that it could be a source as abalone bait for higher protein.There were no evident differences between the new strain and other materials of G.lemaneiformisin the rates of photosynthetic and respiration.In addition,as the new strain was morphologically similar to cultivar,RSAP finger printing was compared among four strains,specific band of the new strain was screened and converted into sequence characterized amplified regions(SCAR)molecular markers with high stability and specificity to identify the new strain.
