[1] Morphometry of subcellular fractions

1974
Publisher Summary In the present chapter emphasis is placed on methods for morphological analysis of particle suspensions on a quantitative basis. However, because the procedure described yields representative electron micrographs of the sample, it will increase the reliability of the morphological examination and reduce the time necessary to perform such analysis, even when only a qualitative evaluation is sought. In the method presented in this chapter, thin pellicles are obtained by filtration on Millipore filters. As originally described the apparatus consists of a special filtration chamber, provided with an automatic control. Although such a system should be preferred when samples are treated routinely, a description of an apparatus based on components which all are commercially available is given in the chapter. Automatic control is indeed convenient but not essential for the method.
