Screening the Lipid-lowering Active Constituents of Herba Cistanches by Yeast Two-hybrid Assay

Objective To screen the lipid-lowering active constituents of Herba Cistanches by yeast two-hybrid assay.Methods Five kinds of constituents from Herba Cistanche were added into YPDA medium with yeast stain AH109 which carried apolipoprotein AI(apo-AI) and scavenger receptors class B type(SR-BI) genes of rat,and their impacts on the interaction of apo-AI and SR-BI were examined by comparing the activity of β-galactosidase.Results Compared with the blank control,allantoin increased β-galactosidase activity,8-epiloganic acid,echinacoside and tubuloside B reduced β-galactosidase activity,and 8-hydroxygeraniol 1-β-D-glucopyranoside had no effect on β-galactosidase activity.Conclusion Allantoin can significantly enhance the interaction of apo-AI and SR-BI,and may have certain lipid-lowering activity.
