Method for titrating rabies viruses quickly with cell plaques

2010
The invention provides a method for titrating rabies viruses quickly with cell plaques, which comprises the following steps of: (1) preparing methyl cellulose solution; (2) preparing cell suspension; (3) diluting rabies virus solution to be measured to prepare rabies virus solution at a gradient concentration; (4) adding the cell suspension into a cell culture plate; (5) adding the rabies virus solution at the gradient concentration into the cell culture plate obtained by the step (4), setting isotype control simultaneously, and putting the cell culture plate into a CO2 incubator for culture; (6) removing supernate in the cell culture plate, adding the methyl cellulose solution into culture pores, and putting the methyl cellulose solution into the CO2 incubator for continuous culture; (7) dying crystal violet and observing the number of plaques in each culture pore of the cell culture plate; and (8) calculating viral titer. The titration method has the advantages of shortening the detection period of the viral titer of the rabies viruses along with simple operation, quickness, accuracy, strong controllability and little lot error.
