Study of paracetamol effect on kidney functional status under experimental insulin resistance in rats

Topicality. Paracetamol (PA) is widely used as an antipyretic and analgesic agent, but the peculiarities of PA effect on organs and tissues under insulin resistance (IR) have not been studied enough. Aim. To study paracetamol effect on kidney functional status under IR in rats. Materials and methods. The study was conducted on white random-bred male rats, which were divided into groups depending on the purpose of the experiment. The IR was modeled by a high-calorie diet enriched with fructose; PA was administered intraperitoneally at a dose of 600 mg/kg body weight once 24 hours before the experiment finishing. The activity of alkaline phosphatase (APh), γ-glutamyltranspeptidase (GGT), and content of total protein, albumins, creatinine, urea, sodium, potassium and chlorides were determined in blood serum. In the urine, the activity of neutral alpha-glucosidase (NAG) and aldolase were determined. Results and discussion. In animals with experimental IR PA administration was accompanied by significant changes in biochemical parameters that reflect the kidney functional status. In particular, an increase in the total protein, albumin, creatinine, urea and electrolytes in the blood serum were significant. Also, the PA injection under IR increased the activity of enzymes, which were investigated in blood serum – APh, GGT, and urine – NAG and aldolase. Conclusions . The obtained data proved the kidney damage development  caused by IR PA administration, in particular, tubular disorders.
