Comparative study between conical column and cylindrical column for preparative liquid chromatography

the flow profiles of solute bands in the conical column with larger inside diameter of inlet than outlet and cylindrical column were evaluated. by an on-column visualization method. in conical column, the band migrated slower in the central region than that in the wall region, resulting in a reverse parabolic flow profile contrast to that in cylindrical column. compared with the cylindrical column with the same volume-and the same length, the conical column had double sample capacity, 36% higher column efficiency, and 12% higher peak height.
