bjective To study biological affection of cmyc antisense RNA on renal cell carcinoma cell line(GRC1) in vitro.ZMethods Based on genetic recombination techniques,an eukaryotic expression vector with cmyc antisense RNA was constructed and the recombinant was then transfected into GRC1 cell line by lipofectin. ResultsBZThe growth rate and colony formation effeciency of antisense RNA transfected cells were much suppressed. The cell cycle analysis showed that the S phase fraction was significantly lower and the cell priiferation stagnated in G0/G1 phase.The rate of DNA synthesis was reduced as shown by reduced relative 3 HTdR incorporation,the mRNA transcription of cmyc being also downregulated greatly in transfected cells.In addition,under electromicroscope, there were also many obvious morphological changes occurred in transfected cells. Conclusions cmyc antisense RNA do effectively inhibit GRC1 cell growth and reverse the malignant phenotype of tumor cell in vitro.
