The Effect of Rofecoxib on Endometriosis in Rats

Objective:To investigate the effects of Rofecoxib in endometriosis model rats.Method: The rat endometriosis model was produced by autotransplantation with uterine tissues.4 weeks after the autotransplantation,rats were divided randomly into three groups.One was treated with Rofecoxib(30 mg/kg) through intragastric administration every day for the four consecutive weeks.Nemestran and saline solution were taken as positive and negative controls respectively,the size of the autotransplants and histopathological condition were examined.The expressions of vascular endometrial growth factor(VEGF) and cyclooxygenase-2(COX-2) in autotransplants were detected through immunohistochemistry.Result: After 4 weeks treatment,the size of the autotransplants of treated groups was not significantly reduced compared with the control group(P0.05).However,histopathological examination showed that the growth and cell proliferation of endometrium were marked repressed,or degeneration and necrosis occurred in most of the cases treated with Rofecoxib and as well as with Nemestran.Moreover,most of the transplanted tissues of treated groups were short of blood supply.There were no significant change in the expressions of VEGF and COX-2 in autotransplants among all groups(P0.05).Conclusion: Rofecoxib causes repression of ectopic endometrial growth and cell proliferation,and degeneration and necrosis of transplanted endometrial tissues.Rofecoxib may not affect the expression of VEGF and COX-2 in the autotransplants,However,the mechanism remains uncertain.
