Determination of the Concentration of Cefetamet in Human Plasma by RP-HPLC

OBJECTIVE: To establish a RP-HPLC method for the determination of cefetamet in human plasma.METHODS: Hypersil C18 was used as column,and 0.04% perchloric acid-acetonitrile(83∶ 17) was taken as mobile phase.The column temperature was set at 40℃;The flow rate was 1.0mL· min-1 and the detection wavelength was 265nm.RESULTS: The calibration curve showed good linearity in a concentration range of 0.125~ 8.0μ g· mL-1(r=0.999 8),the minimum detectable concentration of cefetamet in plasma was 0.125μ g· mL-1,The average recovery rate was 100.6%(RSD=1.2%).The within-day RSD was less than 5.3%(n=5) and the inter-day RSD was less than 4.0%(n=5).CONCLUSION: The method is simple,sensitive,accurate and reproducibily and can be applied to the pharmacokinetic and bioavailability study of cefetamet pivoxil.
