Construction of recombinant retroviral vector carrying IL-12 gene and identification of its expression in HeLa cells.

Objective:To package the recombinant retrovirus carrying interleukin 12(IL-12) gene and to observe the expression of IL-12 in cervical carcinoma cell line HeLa.Methods:The retroviral vector pL35P40SN was transferred into retroviral producer cell line PA317 for packaging retrovirus and then the resultant retrovirus was applied to infect the HeLa cells.The integration of IL-12 gene into the HeLa cells genome was assayed by PCR.The expression of IL-12 gene was detected by RT-PCR.Results:The IL-12 gene was integrated into the genome of HeLa cells shown by results of PCR after retroviral infection.The expression of IL-12 gene was confirmed by RT-PCR.Conclusion:The recombinant retrovirus carrying IL-12 gene was packaged successfully and may be a potentially effective strategy for the treatment of cervical cancer.
