Study on Optimization of Conversion of Straw Stalk into Glucose by Trichoderma viride

[Objective] The process parameters for glucose production by Trichoderma viride fermentate straw stalk were optimized based on response surface analysis. [Method] First,3 important factors affecting glucose production,which identified by screening method of Plackett- Burman were time,pH value and straw stalk concentration from 7 factors. Then Box- Behnken design were adopted to further investigate optimization level and mutual interaction between variables of 3 factors. The optimal conditions of glucose production by Trichoderma viride were obtained by response surface methodology. [Result] The experimental results showed that pH velaue and straw stalk consentration had an individual significant influence on glucose production,and the glucose production up to 191.7 mg/g at the optimal conditions that fermentatio time was 2 days,pH value was 4.5 and straw stalk concentration was 2.5%. Comparing with glucose production before optimization,the production yield improved 137.58%. [Conclusion] The study can improve the conversion rate of glucose production by rice straw cellulose,reduce the production cost and can lay good foundation for production of ethanol fuelby fermentation.
