Cloning and ET Induced Expression Analysis of ERF Gene in Gossypium barbadense
2014
Objective: A novel ERF gene( ethylene- responsive factors ERF) was cloned from Gossypium barbadense. Leaves. That is the foundation for the further function study. Method: The total RNA was extracted by RNA Extraction Kit. The full- length gene sequence was obtained using homology- based cloning by RACE( rapid amplification of cDNA ends,RACE) technology. the sequence was analysised with the molecular biology software, and gene expression was analysised under the ethylene induced using q- RT PCR. Result: The full length cDNA sequence is 926bp( GenBank Accession was No. JQ659260). The open reading frame( ORF) is 624bp encoding a residue of 208 amino acids, named GbERFa. The gene expression was highest at 6h under the ethylene induced. Conclusion: The gene involved in ethylene signaling pathways, and may play a certain role in cotton defense reactions.
