The establishment of DNA standard fingerprint in maize variety Huangzao 4 by SSR markers.

The standard DNA fingerprint was an important tool in maize breeding and DUS testing of new variety.The method of building DNA standard fingerprint by SSR marker in Huangzao 4,a famous maize inbred line in China was studied.And 20 pairs core primers for DNA standard fingerprint building were screened out with PIC(polymorphism information content) values of 0.6754-0.903 by which 6-15 alleles were detected.And the characteristical bands of Huangzao 4 were detected by Bnlg125 and Bnlg2162.The cluster analysis with UPGMA from Nei's(1978) divided Huangzao 4 into 3 groups and 3 sub-groups.In addition,8 % polyacrylamide gel electrophoresis systems could detect more bands than 3 % metaphor gel electrophoresis,therefore it could be used for distinguishing maize varieties among close relative groups.The method for building DNA standard fingerprint of maize and DNA sampling were also discussed in this paper.
